We examined whether early-life TL correlates with mortality rates in superb fairy-wrens (Malurus cyaneus) at different life stages: fledgling, juvenile, and adult. Unlike a comparable study on a similar chemical, early-life TL exposure showed no predictive power regarding mortality at any point in the animal's life cycle. Only patients with a substantial likelihood of developing hepatocellular carcinoma (HCC) are eligible for the diagnostic criteria established by the Liver Imaging Reporting and Data System (LI-RADS) and the European Association for the Study of the Liver (EASL) for non-invasive HCC diagnosis. PubMed's database was searched for original research articles, dated between January 2012 and December 2021, that included LI-RADS and EASL diagnostic criteria for contrast-enhanced ultrasound, computed tomography, or MRI. Regarding chronic liver disease, the recorded information for each study encompassed the algorithm's version, the year of publication, the risk status, and the etiologies. Criteria for high-risk populations were scrutinized for adherence, classified as optimal (unwavering adherence), suboptimal (questionable adherence), or inadequate (clear non-compliance). Analyzing 219 initial studies revealed 215 utilizing LI-RADS criteria, 4 using only EASL criteria, and 15 concurrently applying both LI-RADS and EASL criteria. High-risk population criteria were observed to exhibit varying degrees of adherence, with suboptimal, inadequate, or optimal adherence levels seen in 111/215 (51.6%), 86/215 (40.0%), and 18/215 (8.4%) LI-RADS studies, respectively, and 6/19 (31.6%), 5/19 (26.3%), and 8/19 (42.1%) EASL studies, respectively. This discrepancy was statistically significant (p < 0.001), irrespective of the imaging technique utilized. Significant enhancements in adherence to high-risk population criteria were observed based on LI-RADS versions (v2018: 645%; v2017: 458%; v2014: 244%; v20131: 333%; p < 0.0001) and publication year (2020-2021: 625%; 2018-2019: 339%; 2014-2017: 393%; p = 0.0002), demonstrably impacting study outcomes. We sought to determine whether cytolysin-positive Enterococcus faecalis (E. faecalis) could predict mortality in alcohol-associated hepatitis patients, and to assess the protective role of specific chicken immunoglobulin Y (IgY) antibodies against cytolysin, both in vitro and in a microbiota-humanized mouse model of ethanol-induced liver disease.
In a multi-center study of 26 patients with alcohol-associated hepatitis, our findings were consistent with prior results: fecal cytolysin-positive *E. faecalis* was a predictive factor for 180-day mortality in these individuals. Merging this smaller cohort with our previously published multicenter study reveals that fecal cytolysin yields a more effective diagnostic area under the curve, surpasses other accuracy metrics, and boasts a higher odds ratio for predicting death in individuals with alcohol-associated hepatitis, compared to other established liver disease models. Through a hyperimmunization procedure on chickens, we generated IgY antibodies specific to cytolysin, as part of a precision medicine approach. By neutralizing IgY antibodies that recognize cytolysin, the cytolysin-induced cell death in primary mouse hepatocytes was decreased. Ethanol-induced liver disease in gnotobiotic mice, colonized with stool from cytolysin-positive patients with alcohol-associated hepatitis, was lessened by oral administration of IgY antibodies directed against cytolysin.
